What are the latest advancements and future directions in the development of JAK2 inhibitors for myelofibrosis?

I want to mention that I do believe we are in the golden age of novel therapies in myelofibrosis. There are a number of late-stage clinical developments and clinical trials of novel JAK2 inhibitors, as well as other agents that are attempting to inhibit really important pathways outside of the JAK/STAT signaling pathway.

So your specific question, there are a number of JAK2 inhibitors in clinical trials, including type two JAK2 inhibitors that are thought to be much more selective than the traditional JAK2 inhibitors and other JAK2/Rock inhibitors. And novel JAK2 inhibitors explored in China, and this includes a novel therapy called jaktinib.

But outside of the specifically JAK2 inhibitors, there are therapies that are looking at inhibiting BET inhibition or PIM inhibition, or other therapies that are really important in myelofibrosis in combination with JAK inhibitors such as ruxolitinib, momelotinib, and fedratinib.

So in my opinion, I think that we are going to be seeing a lot of therapies on the horizon about how do we either combine a new therapy on top of a JAK inhibitor or add on a new therapy from a JAK inhibitor that wasn't getting the biggest bang that we were hoping for.
