What is fedratinib?

Fedratinib is an additional JAK2 inhibitor. Ruxolitinib inhibits multiple proteins, including JAK1 and JAK2. Fedratinib, on the other hand, has been thought to inhibit more selectively JAK2.

In clinical practice, this is a very specific portion of patients that are likely to benefit from fedratinib specifically.

The JAKARTRA trial and the Freedom trials were looking at the clinical utility of fedratinib, particularly for patients that had a suboptimal response to frontline ruxolitinib. So patients that are typically using fedratinib are those reaching for this therapy in second line after the ruxolitinib therapies weren't working for them.
