Improved Survival with Liso-cel in Relapsed Lymphoma | Manali Kamdar, MD

Hello, my name is Manali Kamdar. I’m an associate professor and the clinical director of the lymphoma program at the University of Colorado Cancer Center.

Today at ASH 2025, I am presenting, on behalf of my coauthors, the four-year follow-up of the long-term study called TRANSFORM.

TRANSFORM is a global, randomized phase three study in patients with relapsed second-line diffuse large B-cell lymphoma (DLBCL). The study compared lisocabtagene maraleucel (liso-cel), a CAR-T cell therapy, versus bone marrow transplant in high-risk patients.

TRANSFORM was a positive study. Liso-cel was significantly more effective and safe compared to bone marrow transplant and is now considered the new standard of care for patients who have relapsed after one line of chemoimmunotherapy for DLBCL.

After completing TRANSFORM, patients treated with liso-cel could enroll in a separate long-term follow-up (LTFU) study. We now present the four-year follow-up of these patients.

TRANSFORM was a randomized study, and patients on the standard-of-care arm could crossover to receive liso-cel upon progression.

The results at four years are very promising. The four-year overall survival for patients who received liso-cel was 62%, with the median not yet reached. The four-year progression-free survival was 52%, suggesting curative potential.

When analyzing timing, patients who received liso-cel in second-line therapy had a 24-month overall survival of 95%, whereas patients who received liso-cel after crossover in third-line therapy had a 25-month overall survival of 78%.

Importantly, no new safety signals or second primary cancers were observed at long-term follow-up.

Overall, at the four-year mark, liso-cel continues to demonstrate high curative intent potential in patients with relapsed DLBCL.
