Improving Outcomes in FLT3-mutant AML: Crenolanib + Chemo | Thomas Cluzeau, MD, PhD

Okay, so hello, my name is Thomas Clouzot from Nice University Hospital in France. So during the hash meeting, it was a pleasure to present the results of a clinical trial. So it was a clinical trial in relapse refractory acute myeloid leukemia mutated for a flea tree. So it was a specific subgroup, a very poor subgroup of patients. Usually we propose at this patient only a treatment with TKI. So this is a normal targeted therapy. And in this clinical trial, we combine a new TKI, so the Crenolanib, combined with salvage chemotherapy and it was a randomized clinical trial. So half of patients receive a Crenolanib plus salvage chemo and half of patients receive salvage chemo alone. This study was positive because we observed an increase in term of event free survival for patients receiving the Crenolanib plus salvage chemotherapy. It was also positive in term of response because there is a significant increase of response rate for the patient receiving the combination. And after we focus in a subgroup of patients, it was more than half of patients, including in the clinical trial. It was patients commutated for another gene, it was NPM1. And the patient commutated for NPM1 and flea tree have a very improvement using the combination with Crenolanib plus salvage chemotherapy. There is an improvement in event free survival, but also in overall survival for this patient and a benefit was better if patient could undergo to allostem cell transplantation and could continue Crenolanib as maintenance after allostem cell transplantation. This is the first study showing in this specific setting of relapse refractory flea tree mutated EML, a benefit to the addition of targeted therapy TKI flea tree targeting, combining to salvage chemotherapy and the drug development will continue with this drug.
