Emerging Treatment for CD30-Positive Lymphoma: A Phase 1 Study | Swetha Thiruvengadam, MD

Hi, I'm Swayetutha Ravangadam from City of Hope, and at this ash, I was honored to present the preliminary data with updated analysis from our phase one first human study evaluating its antibody drug conjugate targeting CD30 with a novel camphotisan derived payload or chemotherapy toxin for patients with relapsed refractory, Hodgkin lymphoma, peripheral tesa lymphoma, as well as diffuse large Bisa lymphoma. So this is a novel antibody drug conjugate, meaning it is delivering chemotherapy toxin directly by recognizing binding to CD30 receptor on the lymphoma cells and then getting internalized and delivering the chemotherapy toxin to these cells. And at the time of analysis, we had 53 patients enrolled to the study. All of these patients were heavily pretreated and had received multiple prior lines of therapy with a median of five prior lines of therapy. And majority of the patients enrolled were Hodgkin with 37 patients with relapsed refractory Hodgkin, the remaining being peripheral tesa lymphoma and diffuse large Bisa lymphoma that received 30 positive. And in the overall cohort of 53 patients, we saw about three quarters, 77% of patients achieving a response with 25% of patients achieving a complete response. The median follow up from the study is short at seven months, so we need longer follow up to better assess durability of these responses. This drug was well tolerated with a very favorable toxicity and safety profile. The most common side effects that were observed from the drug were related to blood counts, as well as skin rashes and as well as gastrointestinal side effects like nausea. Medications were implemented to mitigate the risk, the severity and incidence of nausea. The infections were observed at overall a low rate and neutropenia, a drop in neutrophil count was the most common more high grade adverse event and it was very well managed with growth factor support and did not lead to any treatment discontinuations. So overall, our drug, this drug showed over 90% reduction in tumor size in patients and I think it is very promising and has very preliminary but good efficacy data and good safety data in a very relapsed and refractory Hodgkin patient population as well as shows activity in our T cell lymphoma and DLBCL patients that are CD30 positive.
